To clarify the dynamic metabolizing of proteins in the plant comprehensively, the turn-over rate of amino acids and proteins in mouse-ear cress (Arabidopsis thaliana) were analyzed by the time-courses of the incorporation of 15NH4+
Cultured cells of mouse-ear cress were incubated in Murashige and Skoog medium which added (15NH4)2SO4 for 10 days. The 1.0 g of cells were mixed with a lysis buffer containing and 5% SDS. The extracted proteins were separated by SDS-PAGE. Those proteins were identified comprehensively with peptide mass fingerprinting by trypsin digestion. The turn-over rates of proteins were analyzed by 15N-incorporation of peptides which determined by MALDI-TOF MS. It was shown that amino acids that have similar metabolic pathway have the similar turn-over rates. Moreover, the turn-over rates of proteins related to the amino acid sequence identity between mouse-ear cress proteins and rice (Oryza sativa) proteins.
