Abstract
In our previous study, Arabidopsis 160,000 TSS tags have been prepared by our CT-MPSS method, resulting in identification of peak TSS positions for 10,000 genes (Yamamoto et al, Plant J 60: 350, 2009). In this study, we extended Arabidopsis TSS analysis with the aid of an Illumina DNA sequencer. RNA was extracted from several tissues and also stress-treated tissues, processed into TSS tags by the Oligo-Cap method, and subjected to large-scale sequencing. Sequence data was then selected, mapped to the genome sequence, and purified again. Our TSS tag info now covers 90% of the Arabidopsis genes.
