Abstract
Full-length cDNA (FLcDNA) sequences play essential roles for accurate structural annotations of active transcripts and biological characterization of individual genes. Moreover, FLcDNA clones are indispensable resources to experimentally study precise biological functions of genes. In this analysis, we constructed 12 FLcDNA libraries containing 172,000 clones obtained from a two-rowed malting Japanese barley, Haruna Nijo and determined the complete sequences of 24,783 barley FLcDNAs. From the comparison of our data with publicly available barley FLcDNAs, 17,773 nonredundant FLcDNAs were novel, and 22,651 FLcDNAs were barley representatives. Their characteristics such as nucleotide lengths, completeness of predicted ORFs showed the completeness of the FLcDNAs, and functional categorization base on gene ontology terms was similar to those of rice FLcDNAs. We found that 1,699 FLcDNAs were barley-specific missing in the four monocot genomes, and the sequences were shorter than the conserved sequences. A database for the barley FLcDNAs is under construction.
