Abstract
Human somatic chromosomes were observed with optical, transmission electron and scanning electron microscopes to reveal their fine structures. Chromosomes showed spiral constitutions especially observed with optical microscope after the treatment with special hypotonic solution.
Observing with scanning electron microscope chromosomes were constracted with the fine fibers of 25-30 nm in diameter which had been called as elementary fibrils. High resolution scanning electron microscope revealed the histon core and linker DNA.
We summarized the results obtained in our laboratory and showed a schema of spiral configuration from DNA to chromatid.
