The use of a nicotinamide (10-40 mM)-containing medium has been shown to be effective in reducing fibroblastoid cells in pancreatic endocrine (PE) cell cultures. Maintenance of these cultures in the medium appeared to occur in a dose-dependent manner. It was apparently accompanied by a loss of 3T3-cell cultures as evidenced by effective curtailment of fibroblastoid cell growth. A monolayer-formed of PPE cells, free of fibroblasts, was stimulated by glucose, and other nutrients, such as leucine, arginine and glyceraaldehyde. These results suggested that nicotinamide is a useful agent to reduce fibroblasts from the PPE cell culture.
