Enzyme immunoassay of hepatitis B surface antigen (HBsAg) using β-D-galactosidase

Abstract
An enzyme immunoassay was developed for the detection of the HBsAg using β-Dgalactosidase/antibody conjugate and silicon rods or glass beads as a solid phase. The lower level of the detection was approximately 1ng/ml for HBsAg, and the sensitivity was about the same in EIA and RIA. Three groups were studied for the detection of HBsAg. The results are as follows:
1) Ninety two nurses were studied by EIA and RIA, and all nurses showed negative HBsAg in both methods.
2) In 451 staffs in our university, 11 subjects (1.6%) were positive for HBsAg by EIA and 10 out of 11 were positive by RIA and 5 out of 11 by Au-plate. It seemed interesting that a case out of 11 positive cases by EIA was demonstrated negative by RIA, whose serum was inhibited over 50% by the confirmatory test using anti-HBs.
3) Positivity was found in 39 out of 743 (5.2%) residents in N-town, Nagasaki
