The test tube of medium is plugged with cotton covered with two sheets of paper-inside with sulphric-acid paper and outside with Japanese paper (Nihonshi). When the inoculation in culture medium is finished, cotton plug is drawn out from the tube, and immediately immersed into dissolved paraffin at 100--120°C and plugged again. By using this procedure the quantity of paraffin for sealing is decreased by 1/10 compared with the method used at present, and penetration of paraffin into cotton is inhibited by sulphric acid paper, so that cotton can be used repeatedly. Also saprophyte is prevented from entering into culture medium through cotton plug. Thus culture medium is not contaminated and good result is obtained.
