Determination of the number of tubercle bacilli present in organs is indispensable for a certain field of tuberculosis research activities. For this determination, two methods have been employed, namely, one is the direct counting method by stained samples and the other is the quantitative cultivation method. The latter method is much more popular nowadays. The quantitative cultivation method, however, has some defects such as the difficulty in findingou. the suitable dilution of the emulsion of an organ to he examined or it takes more than a month before knowing the results.
