Abstract
I tried Feulgen Process, HCI-Giemsa method and thionine staining on Escherichia coli, Shigella dysenteriae, Salmonella paratyphi B. Diplococcus pneumoniae, Spreptococcus and Staphlococcus, and I could discern the nuclei of bacteria as Rohinow, Boivin and others informed.
With bacilli I generally noted 1-8 nuclei in one cytoplasm, ; with cocci 1 or 2. Often were observed images which were supposed to he under process of division of nuclei. In a variant, which was, obtained from Shigella dysenteriae through ultraviolet irradiation and which long remains fibrous, I noted many nuclei, and it is considered that only the divisioi of cytoplasm was impeded there.
In comparing Feulgen process, HCI-Giemsa method and thionine staining, the second was found best in that with it I can differentiate protoplasm from nuclei most distinctly.
