The purpose of this paper is to demonstrate the immunological measurement of properdin and its clinical significance. Properdin was measured by single radial immunodiffusion method and relationship between properdin level and CH50, β1A, β1E, β1F and C3A was investigated in 39 samples from the children with acute glomerulonephritis, 32 from vascular purpura and 30 from healthy adults. Reactivity of purified properdin was assayed with immunoelectrophoresis. The range of properdin level in AGN is 0-73.5% of normal human serum and mean value is 43.5±24.3% in acute phase, and 81.56±26.50% in remission respectively. In VP, the range is 28-117% and mean value is 67.08±27.29% in acute phase, 88.40±13.13% in remission. Correlation berween properdin level and each component of complement was studied. High correlation (γ=0.626) was revealed between properdin and β1A in AGN, and no correlation was found in the serum of VP. In 3 of 10 cases of VP with low titer of properdin, decreasing of properdin level was not accompanied with low CH50 but properdin decreased before renal involvement occurred.
