Delivery of exogenous molecules into live cells is one of the most essential techniques in current molecular biology and biomedical applications. Among various techniques, optical injection by the use of ultrashort pulse laser has proven to be an effective approach for targeted molecular delivery and transfection into single cells. Focused ultrashort laser pulse onto a cell membrane through a microscope objective creates a sub-micron temporal pore, and exogenous molecules enter to a cytosol. Recent decades have seen the development of the delivery of exogenous molecules into a single mammalian cell (NIH3T3) and plant cell (Tobacco BY-2) using a femtosecond Ti:Sapphire amplifier. In this review, we demonstrate the delivery of various molecular tools such as DNA plasmids and mRNA for genetic engineering into a targeted single cell in the embryos of various animals is possible.
