Abstract

A quantitative study on paper chromatography of N-glycoside was done using sulfanil-amide-N-D-glucoside as an example. A spot, which was made by N-glycoside on a paper strip was cut out, extracted and decomposed by 1% HCl-acidic 50% methanol for 10min. The aglycon in the extraction was colored by 0.5% dimethylaminobenzaldehyde-H2SO4-acidic 50% methanol. This final solution was tested through the Beckman's spectrophotpmeter with 450 mμ wave length. As the result, quantitative recovery factor obtained for sulfanil-amide-N-D-glycoside in this method was 1.20.
Applying this method, we repeated the same experiment in the formation of N-glycoside as we did in the previous work, but quantitatively this time. It was formed in 10 cc water medium which contained 10-2 moles of both sulfanilamide and glucose or xylose using 0.2 cc of 2 N acetic acid per 10 cc solution as the catalyser at 30°C. The amounts of N-glycosides formed after 12 hrs. were in the forms of sulfanilamide-N-D-glucoside and N-D-xyloside 5.1 and 7.4% respectively.
This method may be appropriate for a micro-determination of the N-glycosides of aromatic amines geaerally, as we showed in a trial of extention to p-aminobenzoine acid. sulfathiazole, and salfadiazine.
