The purpose of this study is to elucidate the neutrophil propulsion mechanism by cytokine. It is generally known that the interactions between cytokines and receptors on the neutrophil surface are related to signaling by calcium ions (Ca2+). In this study, to clarify the effect of calcium ion concentration on the neutrophil propulsion mechanism, observation experiments with a microscopy are conducted. The time history of calcium ion concentration is obtained by Fura-2. As a result, it was found that stimulus by cytokines reduces calcium ion concentrations on the neutrophil surface. It was also found that calcium ions on the surface are consumed in
dead neutrophils, but it doesn’t move. It is concluded that calcium ions have an important role in signal transduction and are consumed by cytokine stimulus.
