We studied relationship between the efficiency of sonolytic hydrolysis of peptides and the roughness of the tip surface of probe. Adrenocorticotropic hormone fragment 18-39 (ACTH18-39; Mr 2465.7) was used as an analyte peptide. Peptide aqueous solution (10pmol/μl) added catechol was irradiated with 20kHz ultrasound at average output power of 15W. The irradiation horn (3.2 mm diameter and 50 mm in length) was polished by sandpapers, and immersed into the sample solution 15 mm below the surface. Irradiation time was 0 to 60 min. The sonochemical products and reaction efficiency were analyzed using a matrix-assisted laser desorption/ionization time-of-flight (MALDI TOF) mass spectrometer.
