Abstract
In order to select a specific substrate for elastase in the GCF (gingival crevicular fluid), the enzyme activities of granular leukocytes, pancrease and GCF were analyzed using commercial elastase substrates, namely, Methoxysuccinyl-Ala-Ala-Pro-Val-p-nitroanilide, Pyr-Pro-Val-p-nitroanilide, Glp-Pro-Val-p-nitroanilide and Boc-D-Phe-Ala-Nle-p-nitroanilide. The experimental results revealed that among the substrates, Pyr-Pro-Val-p-nitroanilide showed the highest specificity for GCF elastase as well as granular leukocyte elastase. In addition, the relative value of the GCF elastase activity for each substrate was almost same as that of granulocyte elastase, with no significant difference between the GCF elastase and granulocyte elastase activity. Therefore, it was confirmed that the elastase activity in GCF was mainly derived from granular leukocyte elastase.
