It is well known that infection of gingival epithelial cells by Porphyromonas gingivalis contributes to the onset of gingivitis. In this study, we demonstrated the effects of hinokitiol and Paeonia albiflora root extract in suppressing P. gingivalis infection.
First, we tested the effects on the gingipain activity in a purified culture supernatant of P. gingivalis. The results revealed that both hinokitiol and Paeonia albiflora root extract inhibited gingipain activity. Then, we demonstrated, using a co-culture model, that Paeonia albiflora root extract inhibits the adhesion of P. gingivalis to Ca9-22 cells. Finally, we found that both hinokitiol and Paeonia albiflora root extract decreased the viable amount of P. gingivalis invading Ca9-22 cells.
These results suggest that hinokitiol reduces the viable amount of P. gingivalis invading the gingival epithelial cells via inhibiting gingipain activity, whereas Paeonia albiflora root extract suppressed P. gingivalis infection via inhibiting the cell adherence activity of P. gingivalis as well as inhibiting gingipain activity.
