Impairment of DNA Synthesis in Homozygous Gunn Rats

Brain DNA synthesis was investigated in Gunn rat with marked cerebellar hypoplasia.
DNA content of jj cerebellum increased linearly during the first 10 days, and leveled off thereafter and was one-third of Jj at 16 days. However, the pattern of increase in DNA content of the non-cerebellar parts of the brain was almost the same in Jj and jj.
The developmental change of 3H-thymidine incorporation into DNA at 2 hours after injection showed characteristic age-dependence in the cerebellum, the non-cerebellar parts of the brain, and the liver. The difference in incorporation rate between Jj and jj cerebellum was noted from 10 days of life and the specific radioactivity of jj cerebellar DNA was 70% at 12 days and 50% at 16 days of that in Jj littermates. By contrast, no difference was found in the non-cerebellar parts of the brain or the liver.
