The flower buds of Arabidopsis were double-fixed, embedded in resin, sectioned into 80 nm thick and observed using transmission electron microscopy (TEM; JEM-1400). Automatic tiling was performed using the software MATLAB and the JEM Toolbox for remote control of the TEM. Automatic image merging was performed by extending the public domain software ImageJ. This wide-range high-resolution TEM image was taken at a magnification of 12,000x and merged using 5,600 images. It shows the stage at which generative cells arose after first pollen mitosis.
