We have reported the cloning of mast cell surface antigen-1 (MASA-1) with an MW of 90 kDa specifically expressed on human mast cells. By immunofluorescence staining MASA-1 was expressed in the lung lesional tissues and the bronchoalveolar lavage fluid obtained from patients with chronic inflammatory diseases such as interstitial pneumonia, bronchiolitis obliterans organizing pneumonia and sarcoidosis. However, the expression of MASA-1 in the human skin tissues was not evident by this method. Thus, specimens of the skin lesional tissues from patients with atopic dermatitis, hypertrophic scar and keloid were immunohistochemically stained with anti-human MASA-1 antibody and goat anti-rabbit immunoglobulin conjugated with peroxidase labeled polymer (the dextran polymer method). The expression of MASA-1 was clearly detected in the skin lesion of atopic dermatitis, hypertrophic scar and keloid, while in the normal skin few mast cells showed the expression of MASA-1. The MASA-1 positive cells corresponded well with the degranulated mast cells. MASA-1 was also expressed on mast cells in the skin lesion in atopic dermatitis of NC/Nga mice. These findings suggest that MASA-1 was ubiquitously expressed on mast cells in the lesional tissues of chronic inflammation. [Jpn J Physiol 55 Suppl:S234 (2005)]
