Hippocampal interneurons are preferential synaptic targets for hippocampal afferents in the early postnatal period and may be important in the proper development of glutamatergic synaptic connections. Here we provide evidences of active long-distance movements of glutamatergic synapses along the dendrites of immature hippocampal interneurons. Movements of postsynaptic PSD-95 clusters were accompanied with simultaneous translocation of presynaptic synaptophysin-positive puncta and were negatively regulated by neuronal activity. Photoactivation of PSD-95-Dronpa indicated integrity of the postsynaptic structures during translocation. The movements were a mixture of actin-dependent rapid oscillatory movements and unidirectional linear movements, which were predominantly retrograde toward the cell body of postsynaptic neurons and also sensitive to the down-regulation of essential mediators of dynein-dependent motility, Lis1 and Ndel1. Our study suggests active roles of interneurons in network rearrangement through long-range dynein-mediated translocation of synaptic junctions in the early postnatal hippocampus.
