Recently, possible involvement of Na-K-2Cl cotransporter (NKCC1) in the generation of slow waves was reported in the mouse small intestine (Wouters et al. Am J Physiol Gastrointest Liver Physiol. 2006 290:G1219-G1227). In the present study effects of inhibitors of NKCC1 on the generation of pacemaker potentials recorded from interstitial cells of Cajal distributed in the myenteric region (ICC-MY) were investigated using intracellular recording. Bumetanide (100μM), an inhibitor of NKCC1, transiently depolarized the membrane and inhibited the amplitude and maximum rate of rise (dV/dtmax) of pacemaker potentials without changing the frequency. The similar results were obtained by application of furosemide (300μM), another inhibitor of NKCC1. These results suggest that NKCC1 is unlikely to be involved in the generation of pacemaker potentials of mouse small intestine. NKCC1 inhibitors decreased the peak level of pacemaker potentials, suggesting the possible contribution of NKCC1 for the maintainance of the intracellular Cl concentration under physiological condition.
