Amplification of cyclin Dl (CCND1) has been frequently found in head and neck squamous cell carcinomas (SCCs) including oral cancer, suggesting that CCND1 amplification may play an important role in the tumorigenesis of oral SCCs and may serve as an independent prognostic indicator. In the present study, to investigate CCND1 amplification in 9 primary tumors (PTs) and matched lymph node metastases (LNMs), fluorescence in situ hybridization (FISH) on fine-needle aspiration biopsies (FNA) were performed. When CCND1 amplification was identified in PTs, this genetic alteration was always determined in the matched LNMs. On the other hand, when cancer cells in PTs did not show CCND1 amplifica tion, no tumor cells in matched LNMs exhibited amplification. From this study, it may be concluded that CCND1 amplification develops in carcinogenesis before lymph node metastases occur and is main tainedduring metastasis.
