Abstract
To clarify the clinical significance of urinary 8-hydroxydeoxyguanosine (8-OHdG), we measured urinary 8-OHdG by enzyme immunoassay in 73 patients with type 2 diabetes mellitus and 29 age-matched control subjects. We also studied the relationship between urinary 8-OHdG and different microproteins.
The mean urinary 8-OHdG in the 73 diabetic patients was 12.8± 5.1μg/g. Ecr, significantly higher thanthat in the 29 control subjects (7.6±4.3, μg/g. Ecr, p<0.001). Urinary 8-OHdG correlated with hemoglobin Aic (r=0.356, p=0.0018). In contrast, urinary 8-OHdG did not correlate significantly with indicators of glomerular injury such as urinary transferrin and type IV collagen (r=-0.144 and r=-0.061) or with indicators of tubulointerstitial injury such as urinary N-acetyl-β-D-glucosaminidase activity andβ 2-microglobulin (r=-0.082 and r=-0.125).
These results indicate that urinary 8-OHdG as an indicator of oxidative DNA damage may be affected by glycemic control rather than the severity of renal involvement withelevated urinary microproteins.
