Abstract

The highly efficient culture (HEC) assay is known as in vitro assay to detect residual undifferentiated pluripotent stem cells (PSCs) which possess tumorigenic potential. The MEASURE multisite experiments confirmed reported detection sensitivity (0.001%) under various conditions of hiPSC cell lines and culture medium/coating substrate. In addition, improved detection sensitivity (0.00002%) was achieved by utilizing magnetic activated cell sorting. These results indicate that HEC assay is highly sensitive and robust assay for detection of residual undifferentiated PSCs.
