Abstract

Transgenic rodent in vivo assay is useful for assessing the risk of combined exposure of multiple chemicals in the environment. We have evaluated in vivo mutagenicity of chemical mixture contained in airborne particulate matters (PM) by using gpt delta transgenic mouse. We showed that the inhalation of diesel exhaust (DE), a model pollutant, elevated the mutation frequency in lung, and mutation spectrum of DE resembled that of 1,6-dinitropyrene. The extract of PM collected in urban area was revealed to induce G-to-T transversion at a specific position on gpt as a landmark of oxidative stress.
