Various in vitro assays have been used in the drug discovery to reduce the risk on drug development due to safety concerns (Loiodice et al., 2019). However, conventional in vitro assays often apply cells in two-dimensional culture conditions, and the assays are performed under conditions that are substantially different from those of cells in the body (Joshi et al., 2018). As a result, low accuracy in predicting the toxicity observed in clinical trials and in vivo toxicity studies is often a problem (Proctor et al., 2017). Microphysiological systems (MPS), which have received much attention in recent years, are expected to enhance the function of cultured cells and improve the accuracy of prediction of drug-induced toxicity by cultivating cells in a microenvironment that mimics physiological aspects in vivo (Kopec et al., 2021). On the other hand, because of the low throughput and difficulty in operation due to its complex usability, MPS may not currently make full use of its inherent potential in the safety evaluation. In this presentation, we will introduce the current situation of MPS in the field of discovery toxicity together with internal examples, and discuss the characteristics required in the future. In addition, we would like to introduce the advantages/disadvantages of MPS and precautions for handling that we felt when we developed a toxicity evaluation system using MPS, from the viewpoint of users of MPS.
