Abstract

Mass spectrometry imaging (MSI) is a visualization tool for the distribution molecules in tissue sections. It is expected to be a new tool in the field of toxicological pathology because it can provide positional and structural information not only on known molecules but also on unknown molecules. Matrix-assisted laser desorption/ionization (MALDI) and desorption electrospray ionization (DESI) are known ionization methods for MSI and are selected according to the purpose and target molecules. DESI eliminates the need for a matrix and complicated preparation. Since the ionization mechanism is similar to the electrospray ionization (ESI) method commonly used in mass spectrometry, low molecular weight compounds can be easily obtained its MSI. In addition, it is non-destructive because no laser is used. Therefore, sections can also be analyzed histopathologically by staining after MSI, leading to additional significant advantages in this field. We have examined the spatial distribution of its components and metabolites in the kidney of rats administered madder color and investigated the relationship between the distribution of carcinogenic causative components, histopathological changes, DNA damage and gene mutations. In this presentation, I will introduce these studies. Furthermore, the present usefulness and future prospects of MSI in the field of safety research will be discussed.
