Abstract

Tissues and organs are precisely regulated by temporally and spatially defined gene expression. Therefore, to accurately understand this mechanism, it is essential to analyze spatial and gene expression information in conjunction. Here, we have developed a method called Photo-Isolation Chemistry (PIC), which allows comprehensive analysis of gene expression information exclusively from photo-irradiated regions on tissue sections. PIC enables highly sensitive and quantitative analysis of gene expression in micro-regions, including subcellular structures of less than 1 μm, as well as regions of various sizes, such as macroscopic areas from mouse embryos to adult mouse hippocampus. Furthermore, it can be applied to unfixed, fixed-frozen sections, as well as paraffin sections, making it widely applicable from biological research to clinical studies such as pathological diagnosis. In this presentation, we introduce various analysis examples using PIC from its principles and discuss the future technical developments of PIC.
