Abstract
The determination of methylmalonic acid (MMA) in rat urine were studied by use of isotachophoresis. This method is based on measurement of potential unit value of each organic acid. The operational systems for determination of MMA by isotachophoresis were devised. The leading electrolyte at pH of 3.20 using 0.01M HCl and β-alanine was selected as the most suitable. Moreover, the counterflow technique makes it possible to obtain a complete separation of MMA in organic acid mixture and rat urine. The recovery of MMA added to rat urine reached to 94~101%, when the urine was pretreated by Dowex AG 3×4 column. This method for determination of MMA in rat urine by isotachophoresis is simple, rapid and quantitative.
