Determination for β-carotene in Human Serum by Internal Standard Method using Column-switching High-performance Liquid Chromatography with Electrochemical Detection

Abstract
The high-performance liquid chromatographic analysis for the determination of β-carotene content in human serum with retinyl myristate as an internal standard was improved with the aid of electrochemical detection in oxidation mode by a short and long column-swiching method with a species of mobile phase. The mobile phase was acetonitrile, 2-propanol, methanol (71 : 20 : 9) including 0.025 M NaClO_4. The inter-day coefficient of variation were 0.202 ± 0.014 μg/ml (n=6). Also the recovery rate of adding 0.04 μg/ml β-carotene was 92%. It is concluded that this method provided the stable chromatogram by a short-repeated interval measurement for detection of β-carotene in serum at the 10^2 nanomolar level.
