Abstract

There are many methods, on how to stain desoxyribose and ribose nucleic acids (DNA and RNA). However, it is very difficult to obtain a constant result, especially, in cells such as pancreatic cells which contains much RNA. The usual methods can not be applied here, and there seems to be much room for discussion concerning its fixation, concentration of stain, staining time, washing after staining, differentiation and dehydration of sections, etc.
The author suggests a new modified method by using methyl green and pyronin for staining nucleic acid in pancreatic cells. The author believes that this method of staining is the best for RNA in the pancreatic cells, because the result gotten from this method of staining was always constant, and the differentiation between RNA and DNA was very clear. Furthermore, the process was not complicated.
