A systematic approach to convert genomics data into biological knowledge based on protein profiling will be described. The strategy relies on a high-throughput method for the recombinant production of non-homologous regions of the proteome. The protein regions are selected by bioinformatical methods. Such protein fragments are individually used to generate and enrich mono-specific antibodies for systematic analysis of protein profiles in different human organs using tissue arrays. Strategies for cloning, production, and purification of protein fragments as well as purification of antibodies will be described. The results suggests that this antibody based affinity proteomics strategy can be used to produce a proteome atlas, describing distribution and expression of proteins in normal tissue as well as different types of diseased tissues.
