Separation of Isoflavones from Korean Soybean

Isoflavones are important phytochemically active substances found in a variety of plants including soybean. A substance such as phytoestrogen or isoflavones in soybeans has analogous effects to estrogen so that it is useful to relieve the menopause symptoms but also has negative effects for infants such as a growing trouble caused by disturbing a hormone system. As a potential anticancer agent, the isoflavones contained in soybean were considered for separation by reversed-phase high-performance liquid chromatography (RP-HPLC). In this work, analytical and preparative HPLC systems were utilized to obtain isoflavones from Korean soybean. The mobile phases were the ternary system of water/acetonitrile/acetic acid. The optimum operating conditions were experimentally determined with the analytical column (C18, 0.46 × 25cm, 5µm) and the column packed with 15µm packing were used. The experimental variables were the gradient conditions and mobile phase composition. The aglycones isoflavones of daidzein, glycitein, and genistein were identified by purified components.
