Development of Extraction Process of Steroid Saponins from Liriope Platyphylla (Maekmoondong)

Liriope platyphylla (Maekmoondong) is a perennial plant grown in Korea. In this study, Maekmoondong was extracted with hot water (80°C, 2 hr, 2 times), 80% aqueous methanol (80°C, 2 hr, 2 times) and 10-90% aqueous ethanol (80°C, 2 hr, 2 times). The extracts were fractionated with chloroform, diethyl ether, n-butanol and water. For hot-water extraction, crushed dry tubers showed maximum yield (0.1%). Among extraction using hot water, methanol solution, ethanol solutions, the maximum extraction level of crude saponins was obtained (0.40%) when 50% ethanol was used. Among ethanol solutions 60% ethanol yielded the highest soluble solid extract (86.4%). Crude saponin extraction process from Maekmoondong for manufacturing beverages is suggested as follows: Tubers of Maekmoondong is dried and crushed in size of 70 mesh. The crushed tubers are placed in an extraction chamber with 50% ethanol (10 times the tuber) at 80°C for 2 hr. The extract is filtered. The residue was extracted one more time. The combined filtered extract is concentrated in a concentration chamber at 50-60°C and 6 cmHg, reaching the final concentration to 40 Brix.
