A variety of microorganisms were screened for the production of Coenzyme A (CoA), and Pseudomonas alkanolytica Pd 192, Alcaligenes marshallii Ah 197 and Achromobacter nucleoacidives Ba 108, which are n-paraffins-assimilators, were selected as the representative strains.
In regard to the CoA content, n-paraffins-assimilators were generally superior to n-paraffins-unassimilators, and bacteria were superior to yeasts. Moreover, cells grown on n-paraffins maintained higher CoA levels than those grown on glucose. The CoA content was changeable during growth. The maximum content in the cells was at the logarithmic phase, but the maximum productivity per broth was obtained at the stationary phase.
Ps. alkanolytica Pd 192, grown for 10hr in a small, homogenizer-type fermentor, accumulated 80mg/liter of CoA in the culture fluid. Coenzyme A was obtained, in a purity of 90%, from this culture in about 13% yield by Dowex column chromatography, charcoal treatment and DEAE-cellulose column chromatography.
