Abstract

Phospholipase D was purified from Streptomyces lydicus by treatment with CM-Toyopearl 650M, column chromatography on DEAE-Toyopearl 650M and Toyopearl HW50F, and by chromatofocusing. The enzyme preparation was electrophoretically homogeneous and the molecular weight of the enzyme was estimated to be 56, 000. Its isoelectric point was around pH 7.4. The enzyme was most active at pH 6.0 and at around 55°C. It was stable between pH 4 and 7, and below 60°C.
