Abstract

An Imaging Spectrogram Analysis (ISA) system for fluorescence distributions in plant leaves was devised. The ISA system was composed of a microscope, an imaging spectrograph, a chilled-CCD camera, and image analysis devices. Image processing on a personal computer was carried out by software we developed with Microsoft Visual Basic 6.0. The spectral axis and the spatial axis in the system were corrected by measuring the spectrum of a standard light source and by measuring the reflected light from a standard ruler graduated on a slide glass in 10 μm, respectively. Then, transverse distributions of UV laser induced fluorescence in Peanut (Arachis hypogaea L.) leaves were measured. The fluorescence spectra on any line in a section of a leaf were examined in detail. It is thought that this device is useful to examine the growth conditions of various plants.
