The present study reports on the functional and morphological changes in the ovaries in relation to the bioassay findings in this organ. 1) HMG (Humegon, Sankyo-Organon) treatment induced maturation of the primordial follicles to the mature follicles. Increased doses of more than 2 IU of HMG also caused formation of corpus luteum. 2) Following treatment with HMG+anti-HCG serum, the LH activity was suppressed. Histologically maturation of the follicles were demonstrable but the corpus luteum formation was not seen. 3) G-6-PDH and 3β-olDH activities were demonstrated in the granulosa, corpora lutea, theca interna and the interstitial cells. 4) Electron microscopic studies: (a) Granulosa cells: Mitochondria with villous cristae and vesicle like endoplasmic reticulum were seen. (b) Theca interna: The characteristic morphological findings were oblong-shaped mitochondria and endoplasmic reticulum. Deposits of fat granules were seen in the blind ends of the endoplasmic reticulum. (c) Lutein cells: The lutein cells were oval in shape and contained vesicular cristae and mitochondria. They were mostly of the smooth endoplasmic reticulum.
