In order to determine human urinary cortisol by high performance liquid chromatography and fluorometry, prepurification of cortisol in human urine by an open column chromatography on Amberlite CG-50 was examined. A fraction containing cortisol and cortisone was obtained from human urine by adsorption on a column of Amberlite CG-50 in the presence of 35 % ammonium sulfate at pH 2.5 and elution with acidic aqueous ethanol. Ethyl acetate extract of the fraction was analyzed using ODS column (Finepak C18) and polystyrene gel column (Shodex RSpak DS-613). Both gave two peaks that had retention time corresponding to that of cortisol and cortisone respectively. The recovery of 1 μg of cortisol and cortisone added to a urine sample was 90 per cent with R. S. D. of 7.0 %(n=5) and 2 ng of cortisol and cortisone could be detected.
