Fundamental and clinical evaluation was made on flucloxacillin, one of isoxazolyl-penicillins, and the following results were obtained.
1) Following a single ora l administration of flucloxacillin 250 m g to healthy adults, the serum level showed the maximum of 1. 28 mcg/ml at one hour and was slightly detectable at six hours, the urinary recovery rate being 23. 6% in the first eight hours. Transfer of the antibiotic to mother's milk was little, and the serum level in foetus was 1/2∼1/4 of the maternal level.
2) The minimum inhibitory concentration (M. I. C. ) of flucloxacil lin was 0. 78 m cg/ml against most (92%) of the pathogenic Staphylococci isolated from patients, which suggested this agent to be effective against resistant Staphylococcus infections.
3) Flucloxacillin (1 g/day) wa s administered orally for two to ten dyas to twenty-two patients with uterine adnexitis, vulvar furuncle, cystitis, angina, wound infection in the abdominal wall and abdominal wall abscess, with its effectiveness noted in fifteen patients.
4) The results obtained with fiucloxacillin in this study were nearly the same as those with dicloxacillin having similar chemical structure to the agent. It is said that higher serum levels are reached with fiucloxacillin than with other usual isoxazolylpenicillin. In this regard, however, further studies remain to be done.
