The relative rate of protein (hormone) synthesis by various cell types in anterior pituitary of the female mature rat has been studied by means of autoradiographic technique. Normal, castrated and estrogen-treated castrated rats were injected with tritiated leucine and their pituitaries were removed and fixed 2 hrs. after injection. Autoradiographs of pituitary sections wereprepared and autoradiographic grains were counted on the film overlying the cytoplasm of individual cells. Castration promoted the uptake rate of H3-ieucine by gonadotrophs, but did not affect the uptake rate of H3-leucine by the other cells. However, the elevated rate of H3-leucine uptake by gonadotrophs of castrated rats was suppressed by estrogen administration.
