Amino acid and carbohydrate analyses and stability examination of the hypocalcemic polypeptide fraction (P-I) separeted from the Pronase digest of saliva-parotin-A were carried out. The contents of amino acids and carbohydrates in P-I were found to be 52.43 and 20.68%, respectively. Of thirteen kinds of amino acids determined, the high contents of glutamic acid and proline, which were found to be 12.32 and 20.59%, respectively, were characteristic. The hypocalcemic and leukocytosis-promoting activities of P-I were eliminated by treatments with heat, HCl, NaOH, urea and nitrous acid and weakened with nitrogen mustard. These chemical properies of P-I were different from those of the purified saliva-parotin-A.
