The spleen is proved by numerous workers to be one of the important organs to produce antibody [Pfeiffer and Marx1), Wassermann2) etc.] and it is believed that its reticulo-endothelium plays a great rôle in this respect.
To examine antibody production from the organ, various methods have hitherto been devised and recommended. For this purpose, however, tissue culture has been proved to be a very suitable object, for by using this we can know very easily and conveniently the antibody production from the tissue.
Concerning investigations on heinagglutinin production from tissue culture, there have already been some works. Bloom3) found that the in vitro cultivated lung-tissue of a rabbit, immunized by pigeon's erythrocytes, produces an antibody which agglutinates the antigen. Tachibana4), examining the cultures of the spleen, bone-marrow, liver and kidney of a rabbit, treated with the dog's or sheep's erythrocytes, proved hemagglutinin production from the liver and the spleen tissue in the case of immunization with sheep's erythrocytes. Misugi5) who has observed hemagglutinin production from the cultivated spleen and lung tissue of a rabbit, which had been immunized with hen's erythrocytes, noticed that there is a certain relationship between the hemagglutinin quantity and the time-interval from the last antigen injection into the animal body to the commencement of the cultivation of the tissue excised from this animal, as well as the days of incubation passed.
Although, as above mentioned, there are a few studies of hemagglutinin production with tissue culture, it is still a problem remaining unsolved, how drugs influence hemagglutinin production from tissue cultures in vitro, and this is the reason why we intend to carry out the present work.
