Abstract

A simple method was developed for the determination of grown mycelial content in rice-koji by the use of a commercial fungal cell wall lytic emzyme. The sample of rice-koji was dried at 100° for 1 hour, pulverized, washed out residual sugars with 50 mM phosphate buffer (pH 7.0), and digested with a fungal cell wall lytic enzyme, Yatalase. Mycelial content was calculated by determing the amount of N-acetylglucosamine (G1cNAc) liberated from the rice-koji. It was proved that this method was more efficient for the estimation of mycelial content in rice-koji, compared with the glucosamine method.
