Understanding neurogenesis is valuable for the treatment of nervous system disorders. To gain more insight into the induction from monkey embryonic stem (ES) cells to neural cells, we sought to identify proteins associated with differentiation from the ES cells into neurons via neural stem cells by Neural Stem Sphere (NSS) method. Protein extracts of the cells in each stage (the ES cells, neural stem cells and neurons) were subjected to two-dimensional gel electrophoresis (2-DE), and identified by peptide mass fingerprinting. We found altered expression of proteins related to axon pathfinding, fatty acid binding protein-related proteins, and some proteins, which were different from those found previously in mouse. Expression levels of mRNA were determined by realtime RT-PCR. Our results provide new insight into the molecular events associated with the differentiation from monkey ES cells into neurons.
