Controls (13 healthy males and 12 females) and 12 gynecological patients (samples taken before, 1 day after and 1 week after laparotomy) were the subjects of this study. Platelets were isolated from EDTA-blood (8mM) by centrifugation. Platelets were washed 3 times with EDTA containing physiological saline (pH 7.4) and suspended in Zeiller and Hannig medium containing 1mM EDTA to a cell concentration of 5×106 cells/ml. The electrophoretic mobility was measured with the Laser Zee system 3000 (Pen Kem Inc., N. Y., USA). The use of this instrument enables the measurement of a large number of cells (approximately 700) within 5min in the stationary layer with the illuminated laser beam. The platelet electrophoretic mobility ranged from -1.357 to -1.549 (mean and SE; -1.451±0.014) and from -1.447 to -1.594 (-1.493±0.011) μm/sec/V/cm in healthy males and females respectively. There was a significant difference between the sexes (P<0.05). Before the operation, the mobility ranged from -1.279 to -1.624 (-1.493±0.029)μm/sec/V/cm in the gynecological patients. One day after the laparotomy, 11 out of the 12 cases showed an increase in platelet mobility with a mean value of -1.555±0.022μm/sec/V/cm. A significant difference in mobility was observed in comparison to the values before the laparotomy (P<0.01). One week after the operation, platelet mobility showed a tendency to return to the preoperative value (-1.515×0.027). This finding suggests that platelets with smaller surface negative charge may be selectively consumed in surgical procedures.
