Background: Chronic gastritis and gastric ulcer are very prevalent digestive tract disease throughout all ages in the world, and are considered to be a global health problem.Usually they are resulting in an imbalance between the protective factors and aggressive factors in the gastric mucosa. H.pylori infection is the most common cause of chronic inflammation of the stomach worldwide. Xiaochaihutang, a classic Chinese herbal prescription, was first recorded in"ShangHanLun"two thousand years ago. To explore efficacy material base and provide modern experimental basis for anti-gastric ulcer of Modified Xiaochaihutang,serum pharmacochemistry of effective positions were studied.
Methods: Analysis of chemical compositions for volatile oil extracts and serum for gastric ulcer model after administration by GC-MS. Analysis of chemical compositions for alkaloids extracts and serum after administration by LC-MS.
Results: For volatile oil positions, a total of 63 compounds were identified in extracts, which accounting for more than 93% of the all volatile oil, including 23.51% α-curcumene, 11.96% zingiberene, 9.56% curzerene, 6.54% β-sesquinene, 4.77% camphene.And 6 prototype components were also detected in serum for gastric ulcer model. For alkaloids positions, a total of 17 compounds were identified in extracts, 6 prototype components and 2 metabolites, as well as the 3/5-O-feruloylquinic acid and palmatine were also obtained in serum.
Conclusions: The research results can definite the effective chemical compositions for anti-gastric ulcer of Modified Xiaochaihutang, also can provide scientific experimental evidences for innovative clinical application of classic Xiaochaihutang.
