In an attempt to elucidate a possible involvement of Xpc gene in the maintenance of genomic DNA in adult organs, we examined spontaneous mutation in brain, liver and spleen of lacZ-transgemic mice at different ages. The three organs showed similar levels of mutant frequency at 2 months of age. The levels were comparable to those of wild type mice. At old age, however, liver and spleen showed increases in mutant frequency and they were higher than those found in wild type mice. Preliminary studies of DNA sequences of the mutants showed that the spectrum of mutation might be different between young and old tissues. The results suggest that the Xpc gene is involved in suppression of mutation in liver and spleen of adult mice. Old brain revealed a slight increase but no difference between Xpc(-/-) and Xpc(+/+).
