Abstract
How polymyxin-B binds to and inactivates endotoxin was studied. Antibacterial property of 30μg of PX was inactivated by 500μg of ET (extracted from S. min R. 595), while the biological activity of 4μg of ET was inactivated completely by 40μg of PX. Then, PX was immobilized on CNBr activated Sepharose by covalent bond. (PX-Seph). 680μg of stabilized PX could remove 40μg of ET by adsorption. Such effect was found also in plasma. Affinity constant (Ka) in this binding showed a high value of 4.06 x 108.
