Abstract
We prepared 5FU- polylactic acid-microcapsule (5FU-PLA-mc). 5FU was microencapsulated with polylactic acid based on the organic phase separation technique. The size of 5FU-PLA-mc was about 200μm in diameter. The rate of 5FU content in capsule was about 40%. The release rate of 5FU from 5FU-PLA-mc was 100% at 50hrs rotating incubation, When 5FU-PLA-mc suspention was injected into the femoral artery of the rabbit implanted VX2 tumor at thigh, it was found that tumor vessels were embolized from smoll artery to large artery on the angiogram. Tumor redction was found on the rabbit injected 5FU-PLA-mc 75mg suspention into the femoral artery, but enormous tumor growing was found on the rabbit of which femoral artery was only ligated and dissected. Polyiactic acrd was inetabolizabie and absorbale, so it was suitable for the drug- carrier material. From the result of experiment, it was suggested that 5FU-PLA-mc was useful in the cancer chemotherapy.
